LASER PHOTOSENSITIZATION AND METABOLIC INHIBITION OF TISSUE CULTURE CELLS TREATED WITH QUINACRINE HYDROCHLORIDE M. W . Berns, S . E1-Kadi, R . S . Olson and D. E . Rounds Department of Zoology, University of Michigan Ann Arbor, Michigan and The Pasadena Foundation for Medical Research Pasadena, California (Received 1 June 1970; in final form 27 July 1970) In a preliminary report (1) it was suggested that nucleolur function could be probed by treatment of tissue culture cells with quinacrine hydrochloride followed by laser microirradiation . Apparently the quinacrine associates selec tively with the nucleolus, thus making this organelle susceptile to selective damage by laser microirradiation . If the laser technique is to be useful for studying nucleolur function, the metabolic effects of the photosensitizing agent (quinacrine) must be elucidated . In addition, since previous studies have suggested shifts ïn absorption characteristics of other amino acridines (2) when bound in vitro, it is important to determine if such shifts occur with quinacrine . This is particularly important in selecting the appropriate wavelengths for microirradiation . In this manuscript we will present data : (a) indicating the quinacrine hydrochloride has only a temporary effect on nucleolur function ; (b) on the in vitro absorption spectrum of quinacrine hydrochloride .

Apparent antimutagenic activity of quinacrine hydrochloride in Detroit-98 human sternal marrow cells grown in culture.
The ability of a small proportion of the cells in cultures of Detroit-98 (D-98) human sternal marrow cells to attach to glass in the presence of 8-azaguanine has been previously shown to be caused by a mutation to resistance to this com pound. This paper presents results of experiments dealing with the effect of the addition of quinacrine hydrochloride to the culture medium of D-98 cells on the...
متن کاملInduction of Chondrogenic Differentiation of Human Adipose-Derived Stem Cells with TGF-β3 in Pellet Culture System
Objective Adult stem cells which are derived from different tissues, with their unique abilities to self-renew and differentiate into various phenotypes have the potential for cell therapy and tissue engineering. Human adipose tissue is an appropriate source of mesenchymal stem cells with wide differentiation potential for tissue engineering research. In this study isolated stem cells from hum...
متن کاملRole of a voltage-sensitive calcium channel blocker on inhibition of apoptosis in sensory neurons of cultured dorsal root ganglia in adult rat
Introduction: Under pathological conditions, abnormal increase in intracellular calcium concentrations is believed to induce cell death. In the present study, a voltage-sensitive calcium channel blocker (loperamide hydrochloride) was used to investigate its role in inhibition of apoptosis in sensory neurons of cultured spinal dorsal root ganglia (DRG). Methods: L5 DRG from adult rats were di...
متن کاملReduction in the frequency of mutation to resistance to cytarabine in L1210 murine leukemic cells by treatment with quinacrine hydrochloride.
The cloning of single cells in agar was employed to study the effect of the addition of quinacrine hydrochloride to cultures of L1210 cells on the frequency of the mutation to resistance to cytarabine (ara-C). In three independent experiments this frequency was reduced 5to 16-fold after prolonged culture in the presence of quinacrine. Attempts to demonstrate a concomitant increase in the number...
متن کاملTHE EFFECT OF QUINACRINE ON THE EXPRESSION OF WNT3A GENE IN MDA-MB 231 AND MCF7 BREAST CANCER CELL LINES
Background & Aims: Triple-negative breast cancer cells refer to any breast cancer that does not express the genes for the estrogen, progesterone, and HER2 receptors. The Wnt signaling pathway is important in the development and progression of various types of cancers. Quinacrine, a derivative of 9-aminoacridine, has been shown to inhibit the growth of several types of cancer cells. In this stud...
